I seek an expert in Molecular Biology of Fungi to join the Plant Pathology group at the ETH Zurich early next year. I am writing to you in hopes that you know an excellent postdoc or exceptional graduate student who might be interested in taking a new position in Switzerland during that time frame, though the starting date is negotiable and I could wait for up to a year for a highly qualified person. I prefer someone who already has finished at least one postdoc. Experience with pathogens or fungi is desirable, but not essential. I will provide a high degree of flexibility and autonomy in research direction and approach to whoever takes this position. The position is renewable annually for up to six years but could be extended further depending on circumstances. The starting salary will depend on qualifications and experience, but typical candidates could expect a starting salary of CHF 80-90,000 ( US$82-94,000 at current exchange rates). Women candidates will be especially welcomed.

We are in an excellent position to do cutting-edge research on the molecular and evolutionary biology of plant pathogenic fungi. We have global collections of 1000s of strains for all three of our favorite model pathogens. Genome sequences are already available and population genomics projects including at least 120 global strains are underway for all three fungi. As a result of our large-scale projects oriented around QTL mapping, RNA seq and population genomics, we have identified many candidate genes that contribute to virulence, fungicide resistance, and thermal adaptation in all three fungi. The successful candidate will lead a research program to determine molecular and cellular functions associated with the candidate genes and proteins.

Zurich is rated every year as one of the most livable cities in the world, and after living here for 14 years I believe it deserves this reputation. The Plant Pathology Group is currently composed of 20 people from 9 countries, and English is the working language of the group. But knowledge of German or French is useful outside of the university.

Over the past years, Vienna has developed into one of the leading centres of population genetics. The Vienna Graduate School of Population Genetics has been founded to provide a training opportunity for PhD students to build on this excellent on site expertise.

Nous disposons d une bourse de thèse pour étudier les mécanismes moléculaires des interactions entre les champignons ectomycorhiziens etles bactéries du sol. La thèse débutera à l automne 2013.

Cette thèse est co-financée par le département EFPA de l ?INRA et l OakRidge National Laboratory (USA) et se déroulera pour partie dans lelaboratoire Interactions arbres-Microorganismes à l ?INRA de Nancy et pour partie dans le laboratoire du Dr. J. Labbé à l OakRidge National Laboratory (Tennessee, USA). Des connaissances pratiques et théoriquesen microbiologie et biologie moléculaire sont souhaitables. L étudiantutilisera des méthodes de biologie moléculaire, microscopie et lesnouvelles technologies à haut débit (génomique, transcriptomique, métabolomique) pour caractériser les mécanismes d interactions entre les champignons ectomycorhiziens, les bactéries du sols et les racines de peuplier.

Connaître les bases de la physiologie végétale pour évaluer l’impact métabolique (métabolome) d’une surexpression d’un gène ou d’une mutation.

Maîtriser les principales techniques de biochimie (dosages de métabolites, activités enzymatiques) et de biologie moléculaire (PCR, qRT-PCR).

Maîtriser l’utilisation des outils statistiques pour le traitement et l’analyse de données biologiques (Excel Stat, R) et de bioinformatique (analyse de séquences, recherche dans les bases de données, génomiques).

Le suivi de cultures en serres et de la récolte d ‘échantillons avec le personnel technique sera également important.

L’Université Gembloux Agro-Bio Tech lance un grand projet à partir d’une plateforme appelée ’agriculture is life’ (www.agricultureislife.be ). Vont être engagés 16 doctorants pendant 4 ans dans le but d’étudier les nouvelles façon de cultiver (moins d’intrant, agroforesterie, culture associées, techniques culturales sans labour, ...). Dans ces recherches, est également prévu un poste pour l’étude d’éliciteurs et un poste pour l’étude des bactéries permettant de préserver la fertilité du sol.

Un poste de demi-ATER sera disponible à compter du 1er septembre 2013 à l’université d’Evry Val d’Essonne. Les activités de recherche se dérouleront au sein de l’URGV (Unité de recherche en génomique végétale) à Evry (91 000).

Profil enseignement Le candidat participera aux enseignements de biologie et physiologie végétale de la Licence mention Biologie (niveau L1, L2 et L3) mais aussi de Licence professionnelle BAIA (Biotechnologies Appliquées aux Industries des Agroressources) ou des enseignements de génomique végétale du master mention « Sciences du génome aux organismes ». Projet de recherche : Etude de protéines de tomate impliquées dans la résistance aux virus

L’étude des mécanismes moléculaires de la résistance aux virus chez les plantes cultivées est une thématique prioritaire de l’INRA dans le cadre du développement d’une agriculture durable et soucieuse de l’environnement. Même si des avancées ont été faites lors des dix dernières années, les événements moléculaires amenant à la résistance ou au développement de la maladie restent encore largement inconnus. Le projet se basera sur l’étude de deux virus de la famille des Potexvirus, le Virus X de la pomme de terre (PVX) et le Pepino Mosaic Virus (PepMV) et de leur impact chez les Solanacées, famille agronomiquement et économiquement très importante (tomate et tabac). Des études antérieures au sein du laboratoire, par double hybride chez la levure, ont permis d’identifier différentes protéines hôtes interagissant avec les protéines virales.

L’implication de ces différentes protéines dans la résistance doit être étudiée de manière plus précise en particulier par des études in planta de sous-expression, sur-expression ou par l’utilisation de la plateforme TILLING de l’URGV (plateforme permettant d’identifier des variants alléliques dans des gènes d’intérêt).

D’autre part nous avons également caractérisé, en collaboration avec le laboratoire de Thierry Candresse, 5 mutants de tomate présentant une résistance altérée vis-à-vis de PVX (Sturbois et al.,

2012). Afin d’identifier les gènes mutés, une stratégie de cartographie couplée aux nouvelles technologies de séquençage a été initiée en collaboration avec le laboratoire de l’EPGV (Etude du Polymorphisme des Génomes Végétaux). Nous sommes actuellement en train d’identifier les 5 gènes mutés. Cette identification nous permettra de découvrir de nouvelles protéines hôtes impliquées dans la résistance aux virus.

L’ensemble de ces études nous permettra d’avoir une idée plus précise des complexes protéiques impliqués dans la résistance aux virus chez les plantes cultivées.

La personne recrutée prendra en charge la poursuite de ces études sur le PVX et le PepMV chez les plantes cultivées en collaboration avec les laboratoires de l’EPGV et de Thierry Candresse (INRA Villenave d’Ornon).
